Stromal Phenoscape and Dual-Targeted Therapy of PIK3CA-driven Congenital Vascular Anomalies

Description of the granted funding

Vascular malformations (VM) are localized lesions originating from abnormal vascular morphogenesis. Clinical consequences of VM can result from aesthetic harm to bleeding, motility disturbances, visual impairment, disfigurement and to disturbation of organ development. No curative treatment exists, and lesion regrowth is frequently seen after surgical excision. Thus, novel treatment approaches are required. The applied project defines disease mechanisms in VM. Role of somatic mutations, commonly found in VM and various cancer types, in modifying cells, lesion microenvironment and vascular growth are assessed. Precision medicine is developed based on the knowledge of lesional heterogeneity by using novel drug targets and gene-based medicines. Particularly patients with large, complicated or life-threatening VM benefit from the research.
